That's a standard retention arrangement. Yes, the screw must be removed.
Pads slide out to the rear - sometimes they are wedged very tight or are
impacted with dirt and debris.

Make sure you have the correct tool - if the tool has slipped and rounds
out the hardware you are screwed (pun intended).
Often they will be assembled with threat locker - apply heat to loosen
(soldering iron or micro torch).
If the hardware is rounded out use a Dremel tool to cut a slot in the
head and use a regular screw driver.
