Wheel (mainly rear wheels) are always challenge for us heavies. My current
racing weight is 270, and I have a tendency to really be hard on equipment.
So far, I have had great success with Mavic CPX 33. Also been running the
Mavic Cosmic with good results. The cosmic uses a mavic hub which is a but
loud, but runs smoothly otherwise. These are wheels for training and are
fairly heavy. I just purchased a pair of Rolf Vigor RS wheels for racing
(the RS is the heavy rider version of the wheel). I've heard great things
about them and the price/weight ratio is pretty nice.

I'm looking into both hand-built wheels from separate components (Campy hubs
with Mavic Open Pro rims) as well as pre-built wheelsets like Mavic,
Campagnolo, Velomax, Zipp, and FSA.

I'm interested in hearing feedback/experiences on ride quality and longevity
from existing users.

Wheels need to be Campy 10spd compatible and are intended for a steel,
lugged road bike.

The strongest wheels I have are the first ones I built with CXP33 deep dish
rims (28H) and
butted spokes. The deeper section rims are stronger and butted spokes act as
shock absorbers.
Hand built wheels have more consistent tension, and are stronger. Wheel
building is not a
black art, it just requires patience.

Don't bother with tie and solder or using locktite on the nipples.

I'm 225 (now) and have been building custom wheels for about 27 years, You didn't say if the wheels were going to be only for racing, training, commuting, 'cross or whatever.
Lately I have been using a wheel built on a used, chopped down LX 32H hub ('cause it was cheap) laced to a recycled Mavic CXP21 ('cause it was cheap) with DT Competition spokes 3X on the drive side where it counts and Wheelsmith aero spokes 2X on the non drive where strength is not as important. The front wheel is inherently a lot stronger owing to it's symmetrical nature so you get away with fewer spokes and lighter rims there.
It's been my experience that production and "boutique" wheels often have well thought out design, great components and crappy execution. A carefully and correctly hand built wheel with the identical components of a production wheel will almost always stay straighter, rounder, tighter and last longer. That's why they cost more, too.
